The Port Macquarie region has a rich history dating back to convict times, with John Oxley being the first European explorer to explore the district in 1818, being sent back to in 1819 by Governor Macquarie to survey the port and countryside with the aim of establishing a penal settlement. The region's electoral district is named after Oxley, and the city or Port Macquarie takes its name from Governor Macquarie.

The penal settlement was established in 1821 by Captain Francis Allman, after three ships set sail with 60 convicts who were chosen based on their good behaviour and skills. All three ships were wrecked as they entered the harbour.

Macquarie wanted the settlement to be self-sustaining, and by 1824 convicts were employed in building, agriculture, blacksmithing and teaching. The early agriculture of the region included wheat, tobacco, cotton, vegetable, maize, poultry, pigs and cows.

The penal settlement was dismantled in 1830, when the first hotel was built in the town. The free settlers focused on wine growing and dairy farming, along with horse breeding for the Indian army.

Port Macquarie's population more than doubled between 1966 and 1981 - by which time it was the state's second-fastest growing centre.
